Each vial contains Geobacillus stearothermophilus ATCC 7953 spores (105–106 per vial) with a fluorescence enzyme substrate (α-glucosidase). After a normal steam cycle, the vial is activated and read in a compatible fluorescence reader. The reader measures α-glucosidase activity from surviving spores: a fluorescence signal at 3 hours is a positive result — the load failed, and everything in that load must be re-sterilized. No fluorescence signal means no enzyme activity was detected — a negative result. Either way the answer is available at 3 hours, with no overnight wait.
Provincial dental regulators and RCDSO/PHO expect biological monitoring of every sterilizer; rapid systems let you run and close out monitoring the same day, and many clinics monitor daily when a rapid workflow makes it practical. One 50-pack covers about two months of daily testing for one sterilizer — with same-day results.

How to Read Your 3-Hour Result
| Negative — pass | No fluorescence signal is detected: no enzyme activity from surviving spores. Release the load. |
| Positive — fail | A fluorescence signal is detected at 3 hours: spores survived the cycle. Re-sterilize everything in that load and re-test before releasing instruments. |
| How the reader decides | The reader measures the fluorescence produced by the α-glucosidase enzyme — the call is instrument-made (positive or negative), not a visual judgement. |
| Incubation | 56–60°C in a compatible fluorescence reader; allow the reader to preheat (about 20 minutes) before inserting indicators. |
| Positive control | Run one unexposed indicator in the control channel at the start of each day — the run is only valid if the control reads positive. |
| Timing | Read at 3 hours. The fluorescence method replaces the 24-hour visual read; do not continue incubating for a visual colour read after a fluorescence result. |
