How to Read Autoclave Tape: Understanding Chemical Indicators

You have just pulled a wrapped instrument tray from the autoclave. The stripe on the tape has turned a deep, even brown, so it looks ready for the next patient — right? Not quite. That stripe tells you the pack went through a heat and steam cycle; it does not tell you the instruments inside are sterile.

What Is Autoclave Tape?

Autoclave tape — also called sterilization indicator tape or steam sterilization tape — is a pressure-sensitive adhesive tape with chemical indicator stripes along its length. Clinics apply it to wrapped cassettes, pouches, and textile packs before sterilization.

Technically, it is a Type 1 process indicator under ISO 11140-1, the international standard for chemical indicators in steam sterilization. Type 1 indicators sit on the outside of a pack and show that the pack was exposed to the process.

How Chemical Indicator Tape Works

The stripes contain a chemical ink that reacts to heat and moisture. In a steam sterilizer, temperatures typically reach 121°C (250°F) or 134°C (273°F) under pressure, and the ink shifts from a light shade to a dark one when it meets that combination.

The shift is a pass-or-fail signal at the pack level. No change means the pack never reached the conditions needed for processing — re-wrap and run again. A change means the outside of the pack saw steam at temperature.

What a Color Change Does — and Does Not — Prove

This is where reading autoclave tape correctly matters most for infection control. The color change confirms exposure to heat and steam; it does not confirm sterilization. A dark stripe cannot tell you whether steam penetrated to the centre of the pack, whether the load was packed correctly, or whether the sterilizer held temperature long enough.

Think of the tape as a "this pack went through the sterilizer" label, not a certificate of sterility. Common Mistakes When Reading Indicator Tape

Even experienced teams slip up at the autoclave bench. Common errors:

  • Judging the color change in poor lighting, or comparing shades across different tape lots.
  • Reading the tape while the pack is still hot and damp, before the indicator has settled.
  • Treating the tape as the only check before releasing a load.
  • Writing off a stripe that looks "almost" changed — a partial change is a fail, not a maybe.

Complementary Monitoring: Internal and Biological Indicators

External tape is only the first layer. Inside each pack, use internal chemical indicators: Type 4 multi-parameter indicators react to two or more critical variables, Type 5 integrators respond to all of them, and Type 6 emulating indicators are formulated for specific cycles. They confirm steam reached the pack's centre.

The biological indicator (BI) goes further: spores of Geobacillus stearothermophilus, the organism most resistant to steam. A negative result is the closest thing to proof that your sterilizer killed microorganisms. Canadian reprocessing standards, including CSA Z314 and Health Canada guidance for dental offices, require routine BI testing on a schedule set by your sterilizer manufacturer and provincial regulator.

Spore testing catches what tape cannot: a failing heating element, an air leak, a cycle drifting out of parameters. If your clinic does not run BIs on a fixed schedule, that is the highest-value change you can make.

  • Place indicator tape on every pack and a Type 4 or Type 5 indicator inside each pack.
  • Read the stripe in good light, after the pack has cooled, and before it leaves the sterilizer room.

  • Run biological indicators on your required schedule.
  • Store tape rolls in a cool, dry area, away from chemicals and sunlight, and check expiry dates.
  • Quarantine and re-process any load with a failed stripe or a positive BI, and investigate before the next cycle.
